I am new seller, first time i response a buyer order on.  Unfortunately i response  after 21 hours after. My account will any problem?

Your response time is an average. So, yes, you will not have an ideal response rate until you receive more messages and respond to them more quickly. Try to respond to future messages as soon as possible (ideally, within an hour) and eventually, your rate will improve.
